Unlock the Fartkour skill by completing the Alternate Universes Collide Again quest for Kyle
To unlock the Fartkour skill, you must complete the "Alternate Universes Collide Again" quest in South Park: The Fractured But Whole. This quest begins after completing the CLASSIfied mission, where you exit Jimmy's house and head to Kyle's house, which is at the far right end of the street.
Upon arriving at Kyle's house, enter through the front door and make your way to the backyard, where you will encounter Cousin Kyle and, later, Kyle's Mom. During the battle, Cousin Kyle will use laser abilities that will damage all your characters, so it is advisable to defeat him quickly. Once Cousin Kyle is defeated, Kyle's Mom will join the fight, and she is a much bigger threat due to her fast speed and powerful melee attacks. It is recommended to ignore Cousin Kyle and focus on defeating Kyle's Mom as quickly as possible.
After defeating Kyle's Mom, the battle will end, and you will be rewarded with the Driedel of Vengeance artifact and the Maximum Jersey DNA slot. Fartkour will also be unlocked as a Buddy Power. To use this power, open Inspection Mode to scan the backyard for a pinwheel. Human Kite will then climb onto your back, carrying you via fart-powered jumps to reach high-up places. Use this ability to retrieve the kite stuck in the tree, thus completing the "Alternate Universes Collide Again" quest and unlocking the Fartkour skill.

Find Skeeter's Wine Bar in the top left corner of the map
To find Skeeter's Wine Bar in the top left corner of the map, you must first complete the "Alternate Universes Collide Again" quest for Kyle (the Kite Hero). This unlocks the ability to use Fartkour to reach rooftops and other high places. You will receive this quest early on the second day as you explore South Park.
Once you have the Fartkour skill, make your way to the quest marker for Skeeter's Roof-Aged Cheese. The building is called Skeeter's Wine Bar, and it's located in the top-left corner of the map—it's the last building on the left on the northernmost road.
When you arrive at Skeeter's Wine Bar, look for a little kite on the left side of the building. Interact with it by pressing L2 or LT, depending on your controller type. This will allow you to call upon Kyle's assistance in using Fartkour to reach the roof.
Once you're on the roof, you'll find Skeeter's Roof-Aged Cheese. Be sure to also interact with the pinwheels on the left of the building to fully explore the area and unlock the ability to use Fartkour independently.

Use this power to pause the security system and enter Medicinal Fried Chicken.
